        Metabolic regulation of stem cell identity: role of intracellular branched-chain amino acid/keto acid balance in cancer stem cells
      • Overview of the research

        My research aims to elucidate how intracellular metabolic reprogramming regulates cancer malignancy and stem cell identity. We have shown that the balance between branched-chain amino acids (BCAAs) and branched-chain keto acids (BCKAs) governs the stemness and malignant properties of triple-negative breast cancer (TNBC). We reported that the activity balance between BCAT1 (a BCAA aminotransferase) and the BCKDH complex controls intracellular BCAA/BCKA dynamics and directly regulates cancer stem cell maintenance and malignant transformation. We are also developing single-cell-level metabolic heterogeneity analyses using the BCAA fluorescence biosensor. Our research would contribute to the development of effective therapeutic strategy against the metabolism-dependent cancer.
