        Molecular mechanism of ABC proteins
      • Overview of the research

        ABC (ATP-Binding Cassette) proteins are transmembrane proteins with well-conserved ATP binding domains. In humans, 48 ​​ABC proteins play an important role in maintaining our health. ABC proteins mainly transport lipophilic compounds and are responsible for maintaining our body's defense against xenobiotics as well as maintaining lipid homeostasis. Recently, by studying the three-dimensional structures and function of an MDR1(P-glycoprotein), the most representative ABC protein, we revealed the structural change caused by the binding of ATP to the ATP-binding domain is propagated to the transmembrane domain and the whole molecule by bringing about dynamic structural transformation. We also clarified the ingenious mechanism of the MDR1 molecule that squeezes lipophilic compounds out of cells. Based on the research infrastructure established in MDR1 so far, we are trying to clarify the molecular mechanism of other important human ABC proteins (for example, lipid-transporting ABC proteins) that are still unknown. This study has the potential to provide a blueprint for the development of new therapeutic strategies targeting ABC proteins.
