    • Profile

      1999年からCREST研究員としてピロール-イミダゾール(PI) ポリアミドの研究を開始した。PIポリアミドは、配列特異的なDNAマイナーグルーブ結合性を有しており、ワトソンークリック塩基対を認識することができる人工機能性分子である。

      現在に至るまで、京都大学 大学院理学研究科 化学専攻 生物化学研究室において、杉山 弘 教授と共に、固相合成を活用して様々なDNA塩基配列特異性を有する機能性PIポリアミドの合成開発を進めている。研究内容は、機能性PIポリアミドの基礎化学的な結合特性の評価や、特異的な遺伝子発現制御機能の解析を中心にして、細胞生物学的な臨床応用を目指している。

      • Overview of the research

        現在の研究は、従来のCPI-ビニル複合体からCBI-インドール複合体へと設計を改良したことを契機に、より効率的な配列特異的アルキル化剤の実現を目指し、合成と機能評価を中心に進めている。また、CAG繰返配列やテロメア配列などに対する特定配列の検出を可能にする蛍光性ポリアミドの合成と機能評価、特定配列に対するHDAC阻害能によって遺伝子発現を活性化するPy-Im ポリアミド-SAHA (suberoylanilide hydroxamic acid) 誘導体の合成と評価も並行して進めている。細胞生物学的な応用も検討しており、マウスに対する抗がん活性評価と培養がん細胞に対する遺伝子発現解析を実施している。
