      • Overview of the research

        Up to today, the emphasis of prevention and treatment for bacterial infectious disease has been on the development of antibiotics and vaccines. However, it is difficult to eliminate a specific pathogen by antibiotics because of their properties, and the excessive administration of antibiotics causes the spread of drug-resistant bacteria and dysbiosis. Therefore, it is especially expected to develop novel drugs with a specific effect against each bacterial pathogen.
        The purpose of this study is to establish an effective and large-scale screening system for isolating the functional molecules which can effectively inhibit the interaction between bacterial molecules and host molecules, especially for protein-protein interaction. For this purpose, we develop two screening systems, one is the low molecular weight compounds and the other is nanobodies (VHH), and try to isolate validate candidate substances as seeds for molecular targeted drugs that can prevent and control specific bacterial infection.
        We have already found a low molecular weight compound that showed a strong growth inhibition against Group A Streptococcus (GAS). This compound could inhibit the binding between of Shr, a hemoprotein receptor (iron transporter) of GAS, and the hemoglobin. This compound showed a strong specificity against GAS, and no inhibitory activity against gram-negative bacteria and other gram-positive bacteria. These observations suggest the possibility to develop new drugs targeting specific bacterial species.
        In this application, we focus on two bacterial species, Staphylococcus aureus (becoming increasingly resistant to antibiotics) and GAS (causing serious invasive infections such as necrotizing fasciitis and streptococcal toxic shock syndrome) to find novel growth inhibitory molecules. We precisely characterize the conformation and physical-chemical properties between target bacterial molecules and host molecules based on the crystal structure, and also establish a screening system that can effectively and high-sensitively analyze the binding state between them. The candidate substances are used as lead compounds for chemical modifications or genetic modifications, and the most effective substance is selected for further analysis. We also construct gene knockout mutants or amino acid substitution mutants of putative binding sites for the candidate substances in target bacteria, and evaluate the effect of these candidates for comparing the growth of these parents strains and mutants and the risk for acquisition of drug resistance. Finally, we evaluate toxicity and protective ability of candidate compounds by animal experiments using mice.
        We believe that this study might become a milestone providing a new treatment strategy for multidrug-resistant bacterial infection and intractable infectious diseases.
