      これまでの研究内容

      1. ヒトV型コラーゲン遺伝子のクローニングとEhlers-Danlos症候群における変異の同定

      V型コラーゲンは他の繊維状コラーゲンの径・強度を制御すると考えられていた。そこで、当該cDNAをクローニングし、さらに遺伝子(Exon-Intron)構造を明らかにした。その結果、V型コラーゲンにはN末端非コラーゲン領域が存在し、これが繊維の径・強度に影響を与えるものと考えられた。さらに、皮膚、関節、血管等の脆弱性を示すEhlers-Danlos症候群の患者で、V型コラーゲン遺伝子の転座およびN末端非コラーゲン領域にてExon skipが生じる変異を同定した。


      2. コラーゲンの不溶化に働く酵素の同定

      コラーゲン前駆体は可溶性で、細胞外でC末端非コラーゲン領域が切断され不溶化する。しかし、その切断部位は非特異的分解を受けやすく、切断酵素の同定が困難であった。そこで始めに、当該切断活性を促進する因子PCOLCEをクローニングした。PCOLCEは蛋白間結合に働くCUBドメイン有していた。そこで、同ドメインを有するハエ胚背腹軸形成因子BMP-1を上記切断酵素であると予想し、これを証明した。以上の研究より、長年課題であったコラーゲンの不溶化機構を明らかにした。


      3. 免疫系細胞に発現するレクチンの同定と糖鎖認識に関する研究

      樹状細胞 (dendritic cell, DC)やマクロファージ(macrophage, Mf)は、多様なpattern recognition receptor(PRR)により病原体を認識する。このPRRの一つであるレクチンの機能を解析するために、マウスDC/Mfに発現するランゲリンおよび5個のSIGNRファミリー遺伝子を新規に同定し機能を解析した。続いて、その中のSIGNR1がサルモネラやキャンディダ等を認識し、サイトカイン産生を昂進する事を示した。また、それら病原体表面のSIGNR1認識糖鎖構造を明らかにした。


      4. 病原体認識における異なるPRRの共同作用に関する研究

      実際の免疫細胞による病原体認識は、多様なPRRにより同時に起こる。そこで、SIGNR1を中心に他のPRRとの共同作用を検討した。その結果、SIGNR1が同じくレクチンであるDectin-1や、タイプの異なるtoll-like receptor (TLR)2および4と物理的に会合し、殺菌性酸素種および炎症性サイトカインの産生を昂進する事を見出した。


      5. レクチンを介した免疫抑制機構に関する研究

      C. albicans表面の可溶性糖蛋白はヒトを免疫抑制状態に導く。そこでヒトレクチンDC-SIGNの同機構への関与を検討した。先ず、糖鎖アレイ解析でDC-SIGNは病原性C. albicans N-glycanには結合するが、非病原性のS. cerevisiae のそれには結合しないことを示した。また、DC-SIGN発現RAW264.7細胞株は、S. cerevisiaeよりもC. albicansのN-glycanに対して多くの免疫抑制性サイトカインIL-10を産生した。よって、C. albicansは当該N-glycanおよび宿主DC-SIGNを介して免疫応答を抑制すると思われる。


      6. 微細粒子に対するサイズ依存的な細胞応答経路の解析

      シリカやアスベスト等の微細粒子はDC/MfからのIL-1bの産生を誘導する。しかし、これ迄は被検粒子が様々サイズの混合物であった為、粒子のサイズに依存する細胞応答が存在するかは不明であった。そこで、骨髄由来Mfを様々な大きさのラッテクスビーズ(LxB)で刺激したところ、1,000および20 nm径LxBはIL-1bの産生を誘導するが、100 nm径LxBはしないことを見出した。さらに1,000 nm径LxBはミトコンドリアからの活性酸素種依存的、20 nm径LxBは食胞からのcathepsin Bの漏出依存的に、独立してIL-1b産生に働くことが示された。これらの結果は、粒子サイズ依存的な応答経路の存在を示し、また、微細粒子の生体毒性機構の解明と新たな治療法の開発に寄与するものと考える。

        Development of new methods to control immune reactions though understanding immune system.
      • Overview of the research

        Our interest is the induction and control of immunity. We focus on dendritic cells (DC), which are a primary antigen-presenting cell in the immune system. We are especially interested in functions of lectin molecules expressed on DC and its relative, macrophage, that recognize polysaccharides on pathogenic agents. The study includes analyses of interaction between polysaccharides and lectins, and subsequent cellular and systemic responses in co-operation with TLR signaling. In this study, we found that certain lectin-polysaccharide interaction induced immune suppressive environment, ameliorating excessive and lethal inflammation. By these studies, we would like to develop new methods to control immune system.
